De novo mini-binder campaign against the VEGFR2 D2–D3 VEGF-binding epitope (PDB 2X1W chain L) — overlapping the clinically-validated ramucirumab site, with direct VEGF-blocking upside. Designs come from a folded de novo pipeline (RFdiffusion3 → LigandMPNN → Boltz-2 → MD), hotspots anchored on the Leppänen 2010 interface. Phase 1 wet-lab at Adaptyv: a ramucirumab (IMC-1121B) calibration block from PDB 3S36 (Fab + scFv + two mutants + negative) anchors the assay; the refined de novo panel runs on the retained target lot. Selection: AF2 pae_interaction (Bennett 2023) + ipSAE (Dunbrack 2025) + physics AND-gates + SolubleMPNN.
